Measure velocity of microbes

  1. Start loggerpro with a new project
  2. Download the movie of ciliates swimming to the desktop.
    1. In loggerpro, select INSERT >> Movie ..., navigate to the desktop, and open ciliate.avi.
    2. Click the red dots in the bottom right of the movie frame. -- this open the controls.
    3. Set yourself on frame 60 ... how many frames per second? might need your glasses for this
    4. Go back to frame zero.
    5. You need to scale the length. I happen to know the the width of the field of view in this microscope at 10x is 0.4 mm. Press the set scale icon, draw a line across the image from one side to the other. Fill in the size to be 0.4 mm. Note that for any movie you make, you need a scale bar. We will show how to insert this in a later class.
    6. Now press the button with a red dot centered on a + sign. This makes the cursor selection tool.
    7. Center the selection tool on the lower black dot (a small ciliate) in the movie and press the mouse button.
    8. You notice each time you do this, the frame advances and you can locate the position of the ciliate on the next frame.
    9. You will also see that the position and velocity of the object are being recorded and plotted.
    10. Discuss the motion of the ciliate, focussing only on vx for now. Is the speed constant, acceleration constant, how many body lengths per second does the ciliate move?
    1. Compare notes on how you are doing the measurements
    2. Think about your errors and the number of significant figures that you will use to record the data
  3. Pick another one of the ciliates. Switch roles and repeat the above steps.
  4. Advance to frame 479, where a much bigger ciliate makes its appearance. Switch roles and repeat the above steps.
